RESUMO

Introduction: Echinoids (sea urchins) provide shelter for a variety of facultative or obligatory ectosymbionts. Objective: To evaluate the hypothesis that decapods and fishes prefer to associate with echinoid individuals and species that have longer spines. Methods: We visually studied the frequency of decapod crustaceans and fishes associated with echinoids in shallow water (< 4 m) and deeper water (5-20 m) at Los Cabos, Baja California Sur, Mexico, during 1-6 January 2019. Results: We inspected 1 058 echinoids of six species. Five decapod species associated with three species of echinoids. When compared with other echinoid species, in shallow water, decapods associated 5.1 times more often with the longest-spined echinoid Diadema mexicanum (7.0 times more decapods per individual D. mexicanum); in deeper water, association frequency was similar for all echinoid species. Fourteen fish species associated with four echinoid species. In shallow water, fishes associated 2.6 times more with D. mexicanum (4.5 times more fishes per individual). There was no preferred echinoid species in deeper water. Longer-spined D. mexicanum had more decapods and fishes. Associations were more frequent in shallow water. Multiple individuals and species of decapods and fish often associated together with a single D. mexicanum. The decapod that presumably is Tuleariocaris holthuisi showed a possible obligatory association with one of the equinoids (D. mexicanum); the other decapods and all fish species are facultative associates. Conclusion: Our results support the hypothesis that decapods and fishes associate most frequently with echinoids with the longest spines, presumably to reduce the risk of predation.

BACKGROUND: The assessment of oocyte quality is, nowadays, a major challenge in aquaculture, oocyte cryopreservation, and environmental science. Oocyte quality is a determining factor in fertilization and embryo development; however, there is still a lack of rapid and sensitive cellular markers for its assessment. Currently, its estimation is pre-dominantly based on morphological analysis, which is subjective and does not consistently reflect the developmental competence of the oocytes. Despite several recent studies investigating molecular markers related to oocyte quality, methods currently available for their determination pose various technical challenges and limitations. In this study, we developed a novel approach based on fluorescence spectroscopy to assess different intrinsic physiological parameters that can be employed to evaluate egg quality in marine invertebrates that are widely used as animal models such as sea urchins and mussels. RESULTS: Different physiological parameters, such as viability, mitochondrial activity, intracellular ROS levels, plasma membrane lipid peroxidation, and intracellular pH, for egg quality evaluation have been successfully assessed in sea urchins and mussels by using specific fluorescent dyes and detecting the fluorescent signals in eggs through fluorescence spectroscopy. CONCLUSIONS: Based on our findings, we propose these physiological markers as useful predictors of egg quality in marine invertebrates; they can be estimated rapidly, selectively, and sensitively by employing this novel approach, which, due to the speed of analysis, the low cost, and easy use can be considered a powerful analytical tool for the egg quality assessment.

RESUMO

Introduction: Sea urchin gonads (roe or uni) are considered a culinary delicacy worldwide. However, only a few species are considered edible and commercialized. The sea urchin Tripneustes depressus has generated the interest of producers in Baja California Sur, Mexico, due to the quality of its gonads. A biological basis for designing a management strategy is key to consider its commercial exploitation. Objective: To determine the reproductive season of T. depressus through description of the gonad stages and reproductive cycle, and to establish its relationship with environmental factors. Methods: We collected monthly samples (October 2016-September 2017), recording in-situ temperature and photoperiod. We evaluated a sample of 1 055 specimens for demographic characteristics, using total weight (g) and test diameter (cm). We also did a histological analysis of gonads from 178 individuals. Results: Average test diameter was 9.70 ± 0.03 cm (5-12.50 cm). Based on the proportion into the gonad of sexual (gametes) and somatic (nutritive phagocytes) cells, we propose five gonad stages (growing, premature, mature, spawning, and intergametic) for both sexes. There were two times of the year when gonads were heaviest and closely corresponded to the growing stage, coinciding with the highest proportions of nutritive phagocytes. Gonad development (growing and premature stages) peaks in the months with the longest daylight periods, with spawning in the shortest daylight periods. Conclusions: Gonad wet weight and adjusted gonad weight are good indicators of the reproductive season of T. depressus. The lowest gonad wet weights were matched the spawning peak in the shortest daylight period (January and March).

Introduction: Photoperiod is, together with temperature and food availability, one of the main stimuli in the regulation of gametogenesis in a wide variety of species. Objective: To evaluate the effect of photoperiod on the production of mature gametes in cultured Arbacia dufresnii. Methods: An experiment was carried out with three varying light-dark regimes/treatments: constant light (24 h light), neutral photoperiod (12 h light, 12 h darkness), and constant darkness (24 h darkness). Twenty females were used in each treatment. All were induced to spawn and, ten randomly selected females from each treatment were induced to spawn again after 30 days. After 60 days, spawning was induced in the remaining females. The gametes were collected in filtered seawater, fixed in Davidson solution, quantified and measured per individual in triplicate in a Sedgewick-Rafter chamber. To determine maturation, fertilization success was evaluated 30 minutes after fertilization. Results: Our results showed that in the aquaculture system, after only two months, mature gametes were obtained, and in the neutral light regime there were 10 times more gametes than the number produced in wild sea urchins during the spawning period in question. We also found that with a greater exposure to light, a lower number of mature gametes was produced. Conclusions: This study suggests the viability of the production of mature gametes in a short period of time as regards Arbacia dufresnii.

Introduction: Sea urchins in the order Spatangoida are the most diverse group of extant echinoids. Objective: Describe a new genus and species of Spatangoida from abyssal depths, and add new records for known species. Methods: Specimens were collected during several cruises at different areas of the southwestern Atlantic Ocean (SWAO), among 37-55° S latitude at depths ranging from 55 to 3 000 m. We present morphological and ultrastructure analyses. Results: Corparva lyrida gen. et sp. nov. (Palaeotropidae) is described from the Mar del Plata Canyon on the Argentine continental slope (2 950 m depth), the first record of this family from Argentina. Corparva gen. nov. differs in having an apical system semi-ethmolytic, and labrum reaching to rear part of second adjacent ambulacral plate. We also report the northernmost distribution and deepest record for Brisaster moseleyi (38° S latitude, 2 212 m depth), the northward extension of the distribution range of Tripylus excavatus (39° S latitude, 74 m depth), and the first record of Abatus philippii and Abatus agassizii at the Burdwood Bank/MPA Namuncurá. Conclusions: The present work brings novel and updated data about the diversity and distribution of spatangoids from the SWAO, including the description of C. lyrida gen. et sp. nov., and new records of species. This shows how much remains to be known about the diversity and distribution of heart urchins in the SWAO, especially from the deep-sea.

RESUMO

Abstract Background: Heat stress proteins are implicated in stabilizing and refolding denatured proteins in vertebrates and invertebrates. Members of the Hsp70 gene family comprise the cognate heat shock protein (Hsc70) and inducible heat shock protein (Hsp70). However, the cDNA sequence and the expression of Hsp70 in the Antarctic sea urchin are unknown. Methods: We amplified and cloned a transcript sequence of 1991 bp from the Antarctic sea urchin Sterechinus neumayeri, experimentally exposed to heat stress (5 and 10 °C for 1, 24 and 48 h). RACE-PCR and qPCR were employed to determine Hsp70 gene expression, while western blot and ELISA methods were used to determine protein expression. Results: The sequence obtained from S. neumayeri showed high identity with Hsp70 members. Several Hsp70 family features were identified in the deduced amino acid sequence and they indicate that the isolated Hsp70 is related to the cognate heat shock protein type. The corresponding 70 kDa protein, called Sn-Hsp70, was immune detected in the coelomocytes and the digestive tract of S. neumayeri using a monospecific polyclonal antibody. We showed that S. neumayeri do not respond to acute heat stress by up-regulation of Sn-Hsp70 at transcript and protein level. Furthermore, the Sn-Hsp70 protein expression was not induced in the digestive tract. Conclusions: Our results provide the first molecular evidence that Sn-Hsp70 is expressed constitutively and is noninduced by heat stress in S. neumayeri.

RESUMO

Background Sea urchins are animals commonly found on the Brazilian shoreline, being Echinometra lucunter the most abundant species. Accidents caused by E. lucunter have been reported as one of the most frequent in Brazil, and are characterized by intense pain and inflammation, consequence of spine puncture in the skin. In order to characterize such toxic effects, we isolated one molecule that caused inflammatory and nociceptive effects. Methods E. lucunter specimens were collected without gender distinction. Spines were removed and molecules were extracted, fractionated by RP-HPLC and assayed for inflammatory and nociceptive activity, in a biological-driven fractionation way, until the obtainment of one active molecule and its subsequent analysis by mass spectrometry (MS and MS/MS). For inflammation, intravital microscopy was performed on the mouse cremaster muscle, in order to evaluate rolled, adherent and migrating leukocytes. Paw edema was also evaluated. For the nociceptive activity, the paw pressure test was performed in rats. Results One molecule could be isolated and related to the inflammatory and nociceptive activity. Regarding inflammation, increase in adherent and migrating cells was observed in the cremaster muscle after the administration of the molecule. Corroborating the inflammatory response, paw edema was also observed, although only in 20% of controls and 20 min after injection. Additionally, this molecule was able to decrease significantly the pain threshold, characterizing hyperalgesia. This molecule was analyzed by mass spectrometry, and according to the exact molecular mass, isotopic distribution and fragmentation profile, it was possible to propose the molecular formula C29H48N3O10. Conclusions One isolated molecule from the spine extract of E. lucunter is able to elicit inflammation and hypernociception in animal models, which is in agreement with the effects observed in sea urchin accidents.(AU)

RESUMO

Sea urchins are marine invertebrates commonly found in coastal areas around the world. The potential mechanisms of injury are primary puncture wounds, envenomation, and the foreign body reaction. A 20-year-old woman presented with multiple, 0.2 to 0.3 cm-sized, erythematous papules with central blackish particles accompanied by numbness and pain on the dorsum of the right foot and ankle for 7 days. She had been injured by a sea urchin 7 days prior at the seaside in Guam. A skin biopsy of the lesion showed inflammation with foreign body reaction. The attending physician removed all the blackish particles and applied mupirocin ointment. Although skin lesions improved over time, numbness and pain still remained. There have been only six reported cases of sea urchin granuloma in Korea, but no reports of sea urchin injury with neurologic symptoms. Herein, we report a rare case of sea urchin injury accompanied by paresthesia.

AbstractThe species Echinometra lucunter, Echinometra viridis, Lytechinus variegatus, Tripneustes ventricosus, and Diadema antillarum are the most common sea urchins of littoral habitats in the Caribbean. T. ventricosus and L. variegatus are associated with seagrass beds, while the other three species usually inhabit hardground substrates. Food preferences of these species are well documented and they are commonly accepted as being primarily herbivorous-omnivorous; nevertheless, few of them have previously been characterized isotopically. We used this approach for assessing the isotopic characterization of five echinoids. We established the trophic position of two groups of co-occurring species and quantified the contribution of food resources in the diet of Echinometra lucunter, considered the most common sea urchin in the Caribbean region. The species T. ventricosus and D. antillarum showed the highest values of δ15N. Sea urchins exhibited similar values of δ13C varying from -11.6 ± 0.63 to -10.4 ± 0.99%. The echinoid E. lucunter displayed the lowest values of carbon, from -15.40 ± 0.76%. Significant differences among species were found for δ15N and δ13C. Seaweed communities exhibited no differences among sites for overall δ15N (F= 1.300, df= 3, p= 0.301), but we found spatial differences for δ13C (F= 7.410, df= 3, p= 0.001). The ellipse-based metrics of niche width analysis found that the hardground biotope species (D. antillarum, E. lucunter, and E. viridis) did not overlap each other. Similar results were obtained for the co-occurring species of the seagrass biotope; however, the distance between these species was closer than that of the hardground biotope species. The Bayesian mixing models run for E. lucunter at all four localities found differences in food resources contribution. The algae D. menstrualis, C. crassa and B. triquetrum dominated in CGD; whereas C. nitens, Gracilaria spp., and D. caribaea represented the main contributor algae to the diet of E. lucunter at LQY. In Culebra Island, no dominance of any particular algae was detected in TMD, where six of the eight species exhibited a similar contribution. Similarities in δ15N between D. antillarum and T. ventricosus may hint towards a similar trophic level for these species, although T. ventricosus is widely accepted as an omnivore, while D. antillarum is considered a generalist herbivore. The lack of overlap among species in the two biotopes seems to indicate a resource partitioning strategy to avoid niche competition among co-occurring species. Rev. Biol. Trop. 64 (2): 837-848. RESUMO

Background Sea urchins can be found throughout the Brazilian coast and are reported to be one of the major causes of marine accidents on the shoreline. Although not lethal, these accidents are reported to be extremely painful. In order to understand the toxinology of the Brazilian urchins, a peptidomic approach was performed aiming to characterize the naturally occurring peptides in both the coelomic fluid and the spine. Methods Animals were collected without gender distinction and samples of the coelomic fluid and spines extracted were analyzed by RP-HPLC and mass spectrometry for peptide de novo sequencing. Results Several peptides were identified either in the coelomic fluid or the spine extract (except for E. lucunter). The peptide sequences were aligned with public deposited sequences and possible functions were inferred. Moreover, some peptides can be cryptides, since their sequences were identified within functional proteins, for example thymosin from Strongylocentrotus purpuratus. Conclusions Although preliminary, the peptidomic approach presented here reports, for the first time, the abundance of novel biological molecules derived from these animals. The discovery of such molecules may be of potential biotechnological application, as described for other organisms; nevertheless, further studies are required.(AU)

RESUMO

AbstractINTRODUCTION:

We present a review of injuries in humans caused by aquatic animals in Brazil using the Information System for Notifiable Diseases [ Sistema de Informação de Agravos de Notificação (SINAN)] database.

METHODS:

A descriptive and retrospective epidemiological study was conducted from 2007 to 2013.

RESULTS:

A total of 4,118 accidents were recorded. Of these accidents, 88.7% (3,651) were caused by venomous species, and 11.3% (467) were caused by poisonous, traumatic or unidentified aquatic animals. Most of the events were injuries by stingrays (69%) and jellyfish (13.1%). The North region was responsible for the majority of reports (66.2%), with a significant emphasis on accidents caused by freshwater stingrays (92.2% or 2,317 cases). In the South region, the region with the second highest number of records (15.7%), jellyfish caused the majority of accidents (83.7% or 452 cases). The Northeastern region, with 12.5% of the records, was notable because almost all accidents were caused by toadfish (95.6% or 174 cases).

CONCLUSIONS:

Although a comparison of different databases has not been performed, the data presented in this study, compared to local and regional surveys, raises the hypothesis of underreporting of accidents. As the SINAN is the official system for the notification of accidents by venomous animals in Brazil, it is imperative that its operation be reviewed and improved, given that effective measures to prevent accidents by venomous animals depend on a reliable database and the ability to accurately report the true conditions.

RESUMO

Abstract The relative size of the Aristotle's lantern of most sea urchins varies in relation to the food availability and it is often used to infer the availability of food of the environment. The aim of our study was to evaluate the lantern morphology plasticity of Arbacia dufresnii in populations with different environmental characteristics, either mussel beds or disturbed areas, by the invasive alga Undaria pinnatifida along the Patagonian Atlantic coast. The test diameter of the sea urchins along with the weight and height of the lanterns, the jaw lenght and rotula lenght were measured. Classic and geometric morphometric analyses were performed to test differences among populations. The length of the jaw was the best indicator of relative changes in size of the lantern. The largest length of the jaw was measured in individuals from disturbed areas dominated by the invasive algae U. pinnatifida. The rotula shape changed with the increase of the diameter of the sea urchins, it tends to be more elongated with larger sea urchins. Rev. Biol. Trop. 63 (Suppl. 2): 339-351. Epub 2015 June 01.

To survive, these echinoderms depend on their immune system, which has developed a number of compounds which act as antimicrobial defense strategies. In this work, the antibacterial and antifungal activities of the extracts of the starfish Oreaster reticulatus and sea urchins Diadema antillarum and Mellita quinquiesperforata collected in the Caribbean Cordobés were evaluated against the bacteria Escherichia coli, Staphylococcus aureus and Klebsiella pneumoniae and against the phytopathogenic fungi Fusarium sp., Scleortium sp., and Rhizoctonia sp. Our results show that O. reticulatus and D. antillarum produce compounds producing bacterial inhibition at low concentrations (< 100 μg / ml), while, M. quinquiesperforata extracts showed no inhibition at any of the concentrations tested. Specifically, the strain of S. aureus was the one with greater susceptibility to the extracts evaluated, i.e. the metabolites produced by echinoderms inhibit more easily gram + bacteria. Only the D. antillarum dichloromethane affected Fusarium sp. with inhibition of 78.2 % and 56.5 % at concentrations of 2 000 and 100 μg / ml. Strains of the fungi Sclerotium sp. and Rhizoctonia sp. had resistance to all extracts. Rev. Biol. Trop. 63 (Suppl. 2): 329-337. Epub 2015 June 01.

RESUMO

Abstract In the Antarctic marine environment, the water temperature is usually between 2 and - 1.9 °C. Consequently, some Antarctic species have lost the capacity to adapt to sudden changes in temperature. The study of the immune response in Antarctic sea urchin (Sterechinus neumayeri) could help us understand the future impacts of global warming on endemic animals in the Antarctic Peninsula. In this study, the Antarctic sea urchins were challenged with lipopolysaccharides and Vibrio alginolitycus. The cellular response was evaluated by the number of coelomocytes and phagocytosis. A significant increase was observed in red sphere cells and total coelomocytes in animals exposed to LPS. A significant rise in phagocytosis in animals stimulated by LPS was also evidenced. Moreover, the gene expression of three immune related genes was measured by qPCR, showing a rapid increase in their expression levels. By contrast, these immune genes showed a depression in their expression by a thermal effect at 5 and 10 °C. In addition, during bacterial injection, the oxygen consumption was higher in challenged animals. Our results showed that the immune response in the Antarctic sea urchin may be affected by acute thermal stress and that this immune response has a metabolic cost. Rev. Biol. Trop. 63 (Suppl. 2): 309-320. Epub 2015 June 01.

Abstract Several species of sea urchins are a potential resource for the production of gonads for consumption in Venezuela. We studied the gametogenic cycle and the gonadosomatic index (GSI) of the rock boring urchin Echinometra lucunter between June 2010 and November 2011 in northeastern Venezuela (Isla Lobos, Península de Araya and Turpialito, Gulf of Cariaco). The gonads are composed of long ramified acini, covered by connective tissue, followed by a thin layer of cells that differentiate into nutritive phagocytes and gametes, which change in their proportion and size with the maturity stage. Generally, several maturity stages were observed in both locations, showing simultaneous spawning. At Isla de Lobos the individuals in reserve stage predominated with a short period of sexual maturity, while mature and spawned individuals predominated in Turpialito. The GSI suggests that the greatest spawning activity is associated to the rainy season; showing a conservative reproductive activity, accruing energetic reserves during the period of higher primary productivity associated to coastal upwelling, when the energy of nutritive phagocytes is transferred to gametes, for gamete development and spawning. The biological and reproductive characteristics of individuals at Turpialito showed higher GSI having more gonadal tissue for commercial consumption. Rev. Biol. Trop. 63 (Suppl. 2): 273-283.

Abstract The high demand for Paracentrotus lividus (Lamarck, 1816) in countries like France, Italy, Turkey, Belgium and Japan, has raised the level of extraction of this echinoderm in recent years and has generated wear urchin populations, which take years to regenerate. The economic importance of P. lividus in Galicia, with an average annual production of 700≤000 kg and a value close to € 2 million in 2013, has led us to perform a variety of experiences in order to analyze the possibilities of the culture of this species in Galicia in raft. We have designed three experiences: 1) Effect of five diets and three densities on the growth of juvenile P. lividus obtained from the natural environment; 2) Growth of juvenile P. lividus obtained from hatchery, fed algae, and 3) Effect of different experimental diets on gonadal index of P. lividus commercial size. The experiments were performed on a USC raft situated in the estuary of Muros-Noia (Galicia) for four years (2007 - 2010), using 2 231 total urchins, from the natural environment (450), hatchery (773) and market size (1 008), being designed various experimental diets that try to increase somatic and gonadal growth in sea urchins, in order to reduce the rise time of juveniles from the wild and hatchery until they reach the size commercial (55 mm diameter), or until they get a suitable size to repopulate overfished areas of the natural environment (20 mm diameter), and the growth of the gonads of sea urchins with different diets. Monthly has taken diameter of juvenile sea urchins in order to determine the effect of different diets on growth and densities of the same, as well as determining the gonadal index (GI) in sea urchins of commercial size to see the effect took different tested diets. With the data obtained estimates that, at present, it would be profitable to feed juvenile sea urchins P. lividus in raft to reach commercial size, due to the low price of this species in the market (3€ / kg). Additonally they could reach optimum size and be used to repopulate areas where natural populations are depleted. This should be combined with the fattening of gonads and extended time of commercial exploitation. Rev. Biol. Trop. 63 (Suppl. 2): 261-272. Epub 2015 June 01.

Abstract There are three main species of regular sea urchins in the Canary Islands. To establish the optimal fishing seasons for two of them, we studied the evolution of the gonadal index in several years and locations, of Paracentrotus lividus (April 2006 to March 2008 on Tenerife island in two locations: Las Galletas and La Jaca and from April 2006 to January 2009 in Gran Canaria island in two locations: Ojos de Garza and Gando), and Diadema africanum (January 2010 to May 2011 on Gran Canaria island in three locations: Risco Verde, Arguineguin and Agaete). In the case of P. lividus, located on the southern edge of their distribution, the presence of several annual maximum gonadal indez peak was observed, probably related to the temperature. These times of gonadal index increases corresponded to the stages of maturation. The main period of maturity coincided with the fall and early winter (August, October and December). There was sporadic emissions of gametes depending on location, in April, during the summer (June, July or August), autumn (September or October) and winter (December, January or February). This fragmented situation is possibly due to exposure to the hydrodynamic area's and food abundance. The maximum presence of D. africanum occurred in the months of May to June, with virtually no variations throughout the year, in the southernmost locality (Arguineguin). In conclusion, the two species of sea urchins would be complementary shellfish resources, as their periods of maximum GI (Gonadal Index, capture time) do not overlap during the year. Rev. Biol. Trop. 63 (Suppl. 2): 251-260. Epub 2015 June 01.

RESUMO

Abstract Lytechinus variegatus (Lamarck) is a common echinoid found on the Atlantic coast of America from North Carolina to southern Brazil and their populations inhabit sub littoral shallow-water environments. This research examined some aspects of reproduction in the southern limit of distribution of the species. A total of 86 adult individuals of L. variegatus (50.8 to 77.0 mm test diameter) were manually collected from September 2000 to September 2001 along the unconsolidated bottoms at the eurihaline sector of Paranaguá Bay (48° 19' W - 25° 35' S) for histological characterization of gametogenesis, sex ratio and gonadal index (GI) evaluation. This population showed a synchronicity in maturation and spawning events (autumn and winter respectively), with a 1:1 sex ratio (χ2 = 2.57, df = 7, p < 0.05). The GI increase was associated with storage of nutritional resources periods (resting stages and proliferation) that occurred between late winter and spring. Biometric analyzes indicated a greater investment in size and/or quantity of gametes between late spring and summer, when it was possible to observe mature sperm occupying on average 63.9 % of the follicular lumen and oocytes reaching values of 70.0 to 75.0 μm. Mature oocyte mean size is the highest diameter of those developed by other regular echinoids in the same latitude. However, the average diameter of mature oocytes of L. variegatus in the southern limit of its distribution was lower than the values recorded for the northern hemisphere. Rev. Biol. Trop. 63 (Suppl. 2): 243-250. Epub 2015 June 01.

Abstract Echinometra lucunter is relatively abundant in the Venezuela coast, however no details are known about its diet. We compare the stomach contents and repletion index of E. lucunter with macroalgae available in their environment (June 2010-August) and the nutrient cycle at the Golfo de Cariaco. Seventy-one species of macroalgae were identified: Rhodophyta (45), Heterokontophyta (11) and Chlorophyta (15). The mean testa diameter was 41.04 ± 6.60 mm. The food preferences included filamentous, siphonal and calcaeous macroalgae; main food items were the filamentous macroalgae Sphacelaria sp. and Herposiphonia sp., and foliose Dictyota sp., including invertebrates such as sponges, bivalves and barnacle crustaceans. The repletion index was 0.8 - 6.0 % (2.6 ± 1.21 %), maximum values were associated to primary productivity cycle, i.e. December 2010 (6.0 ± 2.21 %), April (4.3 ± 0.34 %), June 2011 (4.0 ± 1.79 %). Food preferences could be associated to the upwelling season and productivity cycle. Rev. Biol. Trop. 63 (Suppl. 2): 233-242. Epub 2015 June 01.
